What tirzepatide is

Tirzepatide, developed as LY3298176, is a synthetic 39-amino-acid peptide built on a GIP-analog backbone with a C20 fatty-diacid acylation that extends its half-life. It agonises both the GIP receptor and the GLP-1 receptor from a single molecule.

The original pharmacological characterisation was published by Coskun and colleagues in Molecular Metabolism in 2018; the biased-signalling profile was described by Willard and colleagues in JCI Insight in 2020.

Why it is used as a reference standard

  • It is the best-characterised dual incretin agonist in the public literature, so assay results can be anchored to published values.
  • Its reported imbalance between GIPR and GLP-1R activity makes it a useful probe when separating the contributions of the two receptors.
  • Its reduced β-arrestin recruitment at GLP-1R makes it a standard comparator in biased-agonism assays.
