What retatrutide is
Retatrutide, developed as LY3437943, is a synthetic single-chain peptide engineered from a GIP-analog backbone and reported to act as an agonist at three receptors simultaneously: the glucose-dependent insulinotropic polypeptide receptor (GIPR), the glucagon-like peptide-1 receptor (GLP-1R) and the glucagon receptor (GCGR). Its fatty-acid modification supports an extended half-life relative to native incretins.
The pharmacological characterisation of the molecule was published by Coskun and colleagues in Cell Metabolism in 2022, which reports the in-vitro receptor potency profile and the design rationale for the balance between the three activities.
