Retatrutide, developed as LY3437943, is a synthetic single-chain peptide engineered from a GIP-analog backbone and reported to act as an agonist at three receptors simultaneously: the glucose-dependent insulinotropic polypeptide receptor (GIPR), the glucagon-like peptide-1 receptor (GLP-1R) and the glucagon receptor (GCGR).
